چکیده

rfHE ANNOUNCE\-IENT of the discoveryof vitamin 1312 in 1948 byRickes, et a!.’ in this coumitry amid hi y Smith, et al.2 in Emiglamid imiitiat-e(! its use in various climiical entities. The principal use has been in the treatment of permiicious -anemia. The dosage of this drug has beemi more or less empirically established!. The hematologic amid subjective responses of the patient with permiicious anemia in relapse have been the main fact-om’s in det-ermimiimig dlosage re(luirements. Little is known aboutthe metabolism ofvitamin B12. The effective dosage as determined l)y it-s al)sorption, storage a-mid excretion pat-tem’mi has received relatively little attemition. Chow amid his (0-workers3 have shown that the intramuscular injection of 100 j g of vitamin B12 prod!uced detect-able B12 activity iii the urine of both permiicious anemia amid! nonpermiicious amiemia patiemits. Thus activity was fleeting, lasting approximately twelve hours. The oral admimiistration of 500 ag, on the other hand, resulted in no detectable B12 activity imi the urimie. Further expamisiomi of this work7 resulted in essentially the same results. Chest-emmami, Cuthbertsomi and Pegler4 foumid that t-he amountof vitamin Bm2 excreted imu the urine varied d!irectly with the amoumitgiven. They diefined the relatiomuship betw-eemi the amount givemi amid the amoumit imijectedl by the formula: E = D 1.2D#{176}89,where E is the amoumit of vitamimi B12 excreted after eight hours amid D is the amoumit of vitamin B12 injected intramuscularly. Itis the purpose of this paper to present the urinary excret-iomi pattermi of vitamin B12 imi patients who have been given varying amount-s of vitamin B12 imitramuscularly.

Evidence for intestinal origin of transcobalamin II during vitamin B12 absorption.

The plasma binding of newly absorbed, radioactively labelled vitamin B12 was studied during a urinary excretion (Schilling) test. Vitamin B12, after being absorbed from the gut, enters blood attached to transcobalamin II, which seems to be derived from the ileal enterocyte. The absorbed B12 re-enters the blood stream after the transcobalamin II-B12 complex is cleared by the liver and it is then...

Relationships among urinary aminoimidazolecarboxamide in urine and folate, and vitamin B12 concentrations in serum.

Urinary aminoimidazolecarboxamide (A IC), serum folate, and serum vitamin B12 values were determined in 84 apparently healthy individuals. An automated system for determination of AIC in urine is described. Despite claims to the contrary, we found no evidence of a strong relationship between elevated (e.g., >1.3 zg/mg of creatinine) AIC excretion as reflected in a casual sample of urine and fol...

Vitamin BIZ deficiency and the excretion of ether - soluble acids in the rat

I . Urinary excretion of total ether-soluble acids and of methylmalonic acid was studied in rats on vitamin BI2-deficient diets with and without a vitamin Blz supplement. 2. It was shown that urinary excretion of total ether-soluble acids and methylmalonic acid was increased in vitamin R1,-deficient rats and that this increase was somewhat variable between individual animals, males and females,...

Effect of vitamin B12 derivatives on urinary excretion of methylmalonic acid in liver diseases.

1. Twenty.one patients with liver diseases were studied for their urinary mehylmalonic acid excretion after a valine load by means of an improved thin layer chromatography. 2. Methylmalonic acid positive cases were found in four out of the ten patients with cirrhosis of the liver, all four with cirrhosis and diabetes mellitus, and none with acute hepatitis of icteric phase. No apparent correlat...

Metabolic interrelationship between vitamin B12 and ascorbic acid in pernicious anemia.

S UBNORMAL PLASMA ascorbic acid concentration and rapid plasma ascorbate clearance have been found in patients with vitamin B12 deficiency despite adequate intake of vitamin C. 2 Cox et al.2 noted in their group of vitamin B12-deficient patients that these abnormalities of ascorbate metabolism were still present 7-14 days following therapy with sufficient vitamin B12 to cause clinical and hemat...
